Stronger than guanidine HCl, guanidine thiocyanate is a powerful protein denaturant that is frequently employed to stop RNase activity in the isolation of intact ribonucleic acid. RNase can recover activity after boiling, but is irreversibly inactivated in a 4 M solution of guanidine thiocyanate. When isolating RNA from tissues that are abundant in RNase, such as the liver, these solutions, to which the reducing agent b-mercaptoethanol is frequently added, are employed to inactivate RNAse. In this manner, total nuclear and cytoplasmic RNA can be extracted.
Proteins easily dissolve, cellular structures collapse, and nucleoproteins separate from nucleic acids in the presence of guanidine thiocyanate because protein secondary structure is lost.
Instructions:
This product is soluble in water (6 M), yielding a clear, colorless solution.
Storage/Stability : Filtration through a 0.2 um filter should be used to make solutions sterile.
